Formin’ actin in the nucleus

چکیده

Many if not most proteins can, under certain conditions, change cellular compartments, such as, for example, shuttling from the cytoplasm to the nucleus. Thus, many proteins may exert functions in various and very different subcellular locations, depending on the signaling context. A large amount of actin regulatory proteins has been detected in the mammalian cell nucleus, although their potential roles are much debated and are just beginning to emerge. Recently, members of the formin family of actin nucleators were also reported to dynamically localize to the nuclear environment. Here we discuss our findings that specific diaphanous-related formins can promote nuclear actin assembly in a signal-dependent manner.

DNA damage induces nuclear actin filament assembly by Formin-2 and Spire-1/2 that promotes efficient DNA repair

Actin filaments assemble inside the nucleus in response to multiple cellular perturbations, including heat shock, protein misfolding, integrin engagement, and serum stimulation. We find that DNA damage also generates nuclear actin filaments—detectable by phalloidin and live-cell actin probes—with three characteristic morphologies: (i) long, nucleoplasmic filaments; (ii) short, nucleolus-associa...

Formin’ a Nuclear Protection

In this issue of Cell, Skau et al. show that the formin FMN2 organizes a perinuclear actin cytoskeleton that protects the nucleus and its genomic content of migrating cells squeezing through small spaces.

Filamin A- and formin 2-dependent endocytosis regulates proliferation via the canonical Wnt pathway.

Actin-associated proteins regulate multiple cellular processes, including proliferation and differentiation, but the molecular mechanisms underlying these processes are unclear. Here, we report that the actin-binding protein filamin A (FlnA) physically interacts with the actin-nucleating protein formin 2 (Fmn2). Loss of FlnA and Fmn2 impairs proliferation, thereby generating multiple embryonic ...

Formin' adherens junctions

pithelial cells zipper together thanks to linear actin cables—cables that assemble at newly formed adherens junctions to stabilize them, thereby counteracting retractive forces at sites of cell–cell contact. Now, Agnieszka Kobielak, Amalia Pasolli, and Elaine Fuchs (Rockefeller University, New York, NY) have found that formin-1 drives the actin polymerization that creates the cables. Formin-1 e...

Class II formin targeting to the cell cortex by binding PI(3,5)P2 is essential for polarized growth

Class II formins are key regulators of actin and are essential for polarized plant cell growth. Here, we show that the class II formin N-terminal phosphatase and tensin (PTEN) domain binds phosphoinositide-3,5-bisphosphate (PI(3,5)P(2)). Replacing the PTEN domain with polypeptides of known lipid-binding specificity, we show that PI(3,5)P(2) binding was required for formin-mediated polarized gro...
